1. The literature section gives a short overview on the possibilities of immunotherapy and adoptive immunotherapy of human and canine tumor-bearing patients with special emphasis on the application of interleukin-2 (IL-2) and lymphokine-activated killer cells (LAK).
It was the aim of this study to modify mesenchymal BHK cells by molecular biological means (tet-on expression system) so that these cells produce continuously biological active canine IL-2 (cIL-2). This cIL-2 will be provided for clinical application in an adoptive immunotherapy in tumor-bearing dogs.
2. For this purpose a cDNA for cIL-2 was amplified on basis of isolated and Con Astimulated lymphocytes of a dog according to the publication of Dunham et al. (1995)who had published primers and conditions of an RT-PCR for cIL-2. The cDNA generated under these conditio...
By means of a standardized survey addressed to breeders and owners of Retrievers in Germany, a capture and analysis of data concerning the state of health and life expectancy of the six Retriever breeds (Golden Retriever, Labrador-Retriever, Flat-Coated Retriever, Chesapeake-Bay-Retriever, Curly-Coated Retriever and Nova-Scotia-Duck-Tolling-Retriever) was performed.
Altogether 1708 surveys (= total investigation population) were completed and returned for dogs with a pedigree issued by the Fédération Cynologique Internationale (FCI) and 352 surveys for dogs without an FCI-authorised pedigree. The data from the two groups of dogs were analyzed separately and were compared.
The mean life expectancy was calculated using the data of 263 at time of the survea already dead dogs. The resultant life expectancy was 8,91 years. There was...
The establishment of a clinic specializing in the deformities of lips, jaw, nose and pharynx during the Second World War by one of the founders in this specific field was Wolfgang Rosenthal. He was banned from practising his profession by the Nazis because of his alleged Jewish origin. Only after the verification of his Aryan parentage was it possible for him to sign a contract of lease about the hunting lodge in Thallwitz near Leipzig with the hereditary prince of Reuß – younger line, Gera-Schleiz-Greiz. This paved the way for the foundation of his clinic. After the extension of the rooms for the surgical department was completed, it paved the way for additional fully functioning departments to be opened in the following years. The clinic obtained a worldwide reputation through the scientific evaluation and publication of the outstand...
Lyme disease is caused by the spirochete B. burgdorferi and today is regarded as the most frequent and most important vector-borne illness in the northern hemisphere. The genospecies B. burgdorferi s. s., B. afzelii and B. garinii have long been known as causative agents of Lyme borreliosis, whereas B. spielmanii has only recently been delineated and described as the fourth human pathogenic species.
The objective of this study was to further characterize the antibiotic susceptibility pattern of B. spielmanii, since so far nothing is known about the effects of antibiotic substances against this particular genospecies. First of all, the strains were genotyped applying PFGE for plasmid profile analysis in combination with RFLP analysis and sequencing of the 5S-23S intergenic spacer region. Subsequently, a phylogenetic tree based on th...
Early viral infection elicits potent type I IFN responses and is often associated with lymphopenia, a transient reduction of blood lymphocyte counts, long before the onset of humoral and cellular immunity. We have investigated the direct effect of type I IFN stimulation of lymphocytes on early lymphopenia and on the generation of antibody responses at later stages of infection.
Lymphopenia induced by infection with vesicular stomatitis virus (VSV) or treatment with the Toll-like receptor agonists poly(I:C) and R-848 was critically dependent on type I interferon receptor (IFNAR)-signalling. Using bone marrow-chimeric mice, radio-resistant cells, such as stroma and endothelium, could be excluded as type I interferon targets for the induction of lymphopenia. Instead, adoptive transfer experiments and studies in conditionally gene-tar...
Randomised controlled clinical trial about the effects of hypertonic saline solution on the circulatory system in cows with right displaced abomasum
The objective of this controlled clinical trial was to determine the effectiveness of an infusion of a small volume of hypertonic saline solution compared to an infusion of isotonic saline solution in consideration of the circulatory system in cows with right displaced abomasum.
For that purpose 60 dairy cows (breed distribution: 56 German Holsteins, 4 German Fleckvieh) aged between two and eleven years who were introduced at Giessen’s Clinic for Ruminants and Swine suffering from right displaced abomasum were randomly allocated as test group (n = 30) or control group (n = 30). The treatment was performed either as an intravenous infusion of two liter of hypertonic saline (7,2 % Na...
In this study a television set, a puzzle-feederÔ and prima hedrons (cubical climbing structures) were tested in a laboratory environment for their effectiveness as occupational objects and for their influence on the behaviour of vervet monkeys (Cercopithecus aethiops). The aim of the study was, on the one hand, to satisfy the animal protection law; on the other hand to make a contribution to the physical and psychological well-being of vervet monkeys, which especially in laboratory environment, live under considerable restrictions.
Study animals were two groups of vervet monkeys with six animals per group, both of which have a family structure. Each group consisted of one adult male, two adult females and three juveniles of both sexes. The history and origin of the animals is different from each other. The influence of this on the...
In this study, the molecular mechanisms of the stimulation of hepatitis C virus RNA translation by the liver-specific microRNA-122 were investigated. The hepatitis C virus (HCV) was discovered in 1989 as the main causative agent of the nonA-nonB-hepatitis. HCV infects an estimated 3 % of the world’s population. About 80 % of infected individuals are not able to eliminate the virus. They develop a chronic hepatitis with a high risk for a long-term progress of liver diseases like liver cirrhosis or hepatocellular carcinoma. Since a protective vaccine is not available yet and the therapeutic options are still limited, the detailed understanding of the viral life cycle is absolutely essential. During the last years a positive regulation of the HCV life cycle by the liver-specific microRNA (miR)-122 was discovered. However, the stimulation ...
Regulation of gene expression is very important for proper differentiation and development of higher organisms. At the same time a cell-specific gene expression is crucial for specification of different tissues and organs and thereby for the formation of a multicellular organism. Thus, gene expression in eukaryotic cells is tightly regulated. Insulator elements divide the genome into functional domains by shielding genes from inappropriate enhancer action or by blocking the spread of heterochromatin into transcriptionally active domains. The factor CTCF plays an important role in regulating gene expression. Furthermore, in vertebrates it is the only factor known to execute enhancer blocking activity of insulator elements. However, the mechanism behind this function is still unknown. Moreover, a general participation of CTCF in the barr...
