The microtubule (MT) network is one of the major cytoskeletal systems of all eukaryotic cells, being responsible for a variety of important processes. MT organization is necessary for proper cell division, movement and morphogenesis, among other processes. MT plus-end-tracking proteins (+TIPs) are a diverse group of molecules, which display a dynamic accumulation at the distal ends of growing MTs and help to shape the MT network. In this thesis we examined the functions of EB proteins in controlling MT dynamics and addressed several questions regarding the mechanisms of plus end tracking and the organization of the +TIP network. Our findings allowed us to define better the central role of EB proteins in targeting +TIPs to the growing MT ends and identify molecular mechanisms behind this process
According to Optimum Currency Area literature, the synchronisation of business cycles is necessary for the successful implementation of a common monetary policy. This paper contributes to this debate, providing a descriptive analysis of the euro area business cycle synchronisation, using quarterly real GDP series, spanning from 1980:1 to 2004:4, for nine euro area countries. We consider the four largest countries (Germany, France, Italy and Spain), which account for almost 80% of the euro area GDP, and five among the smallest countries, Belgium, Finland, Greece, the Netherlands and Portugal. Our aim is to see whether the synchronisation and the amplitude of the business cycles across the Member States increased with the monetary integration process. We will give greater focus to small countries compared to what can be seen in existent ...
O doen...
Primary hyperparathyroidism (PHPT) is a common endocrine disorder that mainly affects middle-aged women. Patients are usually asymptomatic. The disease might be ascribable to hyperplasia, carcinoma, and single or multiple adenomas. PHPT may be sporadic or familial, the latter comprising multiple endocrine neoplasia type 1 or 2A, familial benign hypocalciuria hypercalcemia, and hyperparathyroidism-jaw tumor syndrome. The most common causes for persistent PHPT are multiglandular disease, and missed abnormal ectopic or orthotopic parathyroid glands. Imaging localization studies should precede a new surgical intervention. Ectopic parathyroid glands are rarely located at the aortopulmonary window. For diagnosis confirmation, 99mTc-sestamibi SPECT/CT seems to be an advantageous test. Another possibility is to perform 99mTc-sestamibi followed...
Contour-clamped homogeneous electric field pulsed-field gel electrophoresis (CHEF-PFGE) was used to compare Wisconsin isolates of Escherichia coli O157:H7, including 39 isolates from a 1994 day care center outbreak, 28 isolates from 18 individuals from the surrounding geographic area with sporadic cases occurring during the 3 months before the outbreak, and 3 isolates, collected in 1995, from patients with hemolytic-uremic syndrome (HUS) who were from eastern Wisconsin counties other than those inhabited by the day care center and sporadic-case individuals. The technique of CHEF-PFGE using XbaI identified seven highly related restriction endonuclease digestion profiles (REDPs) (93 to 98% similarity) among the 39 day care center isolates and nine XbaI REDPs (63 to 93% similarity) among the 28 isolates from sporadic-case individuals, inc...
ABSTRACT Introduction: Neonates admitted to Neonatal Intensive Care Units (NICU) have a substantial risk for healthcare-associated infections (HAI). A high incidence of HAI was found in Maternidade Júlio Dinis (MJD) NICU and led to changes in daily clinical practice. Objectives: To evaluate the effectiveness of new strategies for prevention of infection implemented in the NICU of MJD. Methods: We conducted a prospective study that included all infants admitted to the NICU of MJD during the 16 months before (group 1) and 16 months after (group 2) the protocol change, comparing the rates of infection in these two time periods. Results: 528 infants were included in group 1 and 593 in group 2, with no statistical evidence of different characteristics between them (weight, gestational age, CVC utilization rate, duration of pa...
Dendritic spines are the major sites of excitatory synaptic input, and their morphological changes have been linked to learning and memory processes. Here, we report that growing microtubule plus ends decorated by the microtubule tip-tracking protein EB3 enter spines and can modulate spine morphology. We describe p140Cap/SNIP, a regulator of Src tyrosine kinase, as an EB3 interacting partner that is predominantly localized to spines and enriched in the postsynaptic density. Inhibition of microtubule dynamics, or knockdown of either EB3 or p140Cap, modulates spine shape via regulation of the actin cytoskeleton. Fluorescence recovery after photobleaching revealed that EB3-binding is required for p140Cap accumulation within spines. In addition, we found that p140Cap interacts with Src substrate and F-actin-binding protein cortactin. We pr...
