[[abstract]]摘要 近年來,堆肥法廣為利用在處理廚餘廢棄物。然而,在堆肥過程中翻動含有許多微生物的堆肥物,會使生物氣膠散佈在空氣中。若吸入生物氣膠微粒,呼吸道表皮細胞會因生物氣膠微粒所含的內毒素與黴菌毒素造成發炎反應。由於生物氣膠微粒中的孢子可深入呼吸道末稍,因此本研究之目的在於定量廚餘堆肥廠生物氣膠中微生物,以及其對人類呼吸道表皮細胞重塑基因(remodeling genes)之影響。 本研究收集廚餘堆肥廠三月至八月的生物氣膠微粒,分別以培養法和即時定量聚合?連鎖反應(real-time quantitative polymerase chain reaction; real-time PCR)對生物氣膠中Aspergillus fumigatus (A. fumigatus)進行定性與定量分析。另一方面,將人類呼吸道上表皮NCI-H292細胞暴露生物氣膠樣本或A. fumigatus標準菌株,以酵素連結免疫吸附分析法(enzyme-linked immunosorbent assay; ELISA)量測發炎前趨激素介白素-6 (interleukin-6; IL-6)分泌量,並以real-time PCR分析重塑基因表現。 研究結果顯示,五月至六月之間堆肥廠中生物氣膠總菌落數最高(4.9104 CFU/m3),其中嗜中溫細菌與麴菌的數量最多,A. fumigatus孢子數也達到最高。生物氣膠樣本誘發NCI-H292細胞分泌IL-6同樣在五月至六月最高(平均值標準差:987129.4 pg/ml),並且與麴菌和嗜中溫細菌之數量呈正相關。在重塑基因表現方面,NCI-H292細胞暴露生物氣膠樣本後造成transforming growth factor (TGF)-b1、epidermal growth factor receptor (EGFR)與cyclin-dependent kinase inhibitor 1A (p21WAF1/CIP1)表...
[[abstract]]雙酚A (bisphenol A, BPA)為塑膠製品中常用的塑化劑,能與雌激素受體鍵結,並為一內分泌干擾物質。BPA為脂溶性,具生物累積性,會囤積在脂肪細胞。脂肪細胞(adipose cells)會分泌脂肪激素(adipokines),如脂瘦素(leptin)和脂聯素(adiponectin),以來維持生物體內能量代謝的恆定性。若BPA累積在脂肪細胞時,可能會影響脂肪激素之調控而誘發代謝性疾病。因此,本研究目的為調查妊娠婦女、不孕症婦女和臍帶血中BPA濃度,以及探討BPA對脂肪激素和雌激素受體基因表現之影響。 由高效能液相層析儀(high-performance liquid chromatography)結合紫外光偵測儀(ultraviolet detector)分析結果發現,妊娠婦女血漿中BPA濃度略高於不孕症婦女,分別為5.44  6.04 ng/ml和5.20  5.30 ng/ml (平均值標準差; p = 0.784)。除此之外,臍帶血中能偵測到BPA (1.06  2.39 ng/ml),表示胎兒會經由胎盤暴露到BPA,並且妊娠婦女血漿中BPA濃度與胎兒體重呈顯著負相關(r = -0.304, p = 0.010)。另外,分別以定量聚合酶鏈鎖反應(real-time polymerase chain reaction)分析組織中脂肪激素和雌激素受體基因表現,及酵素連結免疫法(enzyme-linked immunosorbent assay, ELISA)分析血漿中脂肪激素蛋白質濃度。結果顯示,妊娠婦女血漿中BPA濃度,與胎盤組織之雌激素受體 (r = 0.230, p = 0.048)及雌激素受體 (r = 0.168, p = 0.150)基因表現量呈正相關。此外,妊娠婦女血漿中BPA濃度與血漿中脂瘦素(r = 0.068, p = 0.563)和脂聯素(r = 0.015, p = 0.899)濃度呈正相...
[[abstract]]Statistical static timing analysis (SSTA) is indispensable for nanometer manufacturing under process variability. The process variations cause significant uncertainty in VLSI circuit timing and this makes yield control and timing verification a very difficult challenge. SSTA is suitable for timing estimation and design for manufacturability under process variation. However, most of the existing SSTA techniques have difficulty in keeping closed-form expressions after max operations and sum operations on variation sources. The composting procedure in food waste plants generates airborne bioaerosols that have the potential to damage human airway epithelial cells. Persistent inflammation and repair responses induce airway remodeling and damage to the respiratory system. This study elucidated the expression changes of airway remodeling genes in human lung mucoepidermoid NCI-H292 cells exposed to bioaerosols from a composting plant. Different types of microorganisms were detectable in the composting plant, using the agar culture method. Real-time polymerase chain reaction was used to quantify the level of Aspergillus fumigatus and the profile of remodeling genes. Inorganic arsenic (iAs) is an established transplacental agent known to affect fetal development in animal studies. However, iAs has not been adequately studied in the general population with respect to iAs exposure during pregnancy and its impact on the health status of newborns. The aims of this study were to 1) elucidate the association between arsenic exposure and oxidative/methylated DNA damage in pregnant women, and 2) determine the association with birth outcomes. A birth cohort study of 299 pregnant mother-newborn pairs was recruited during 2001–2002 in Taiwan. We collected maternal urine samples during the 3rd trimester for measuring iAs and its metabolites. In daily life, humans are exposed to the extremely low-frequency electromagnetic fields (ELF-EMFs) generated by electric appliances, and public concern is increasing regarding the biological effects of such exposure. Numerous studies have yielded inconsistent results regarding the biological effects of ELF-EMF exposure. Here we show that ELF-EMFs activate the ATM-Chk2-p21 pathway in HaCaT cells, inhibiting cell proliferation. To present well-founded results, we comprehensively evaluated the biological effects of ELF-EMFs at the transcriptional, protein, and cellular levels.
