In the present work the preparation and reactivity of different α-functionalised organolithium compounds starting from chlorinated substrates is described, involving the metalation with lithium powder, catalised by 4,4’-di-tert-butylbiphenyl (DTBB). In most cases, these compounds are very unstable, for that reason Barbier conditions are required. In the first chapter the preparation of α-oxygenated organolithium compounds is studied. First, the preparation of enantiomerically pure lithiomethyl ethers from chloromethyl ethers is described, which by reaction with electrophiles furnish enantiomerically pure functionalised ethers. Commercially available alcohols are used as chiral source. In the second part of this chapter the preparation of lithiated sinthons of the type HOCH(R)− is described. The homocoupling of terminal alkynes under heterogeneous catalysis is tackled in this perspective. The performance of different unsupported and supported catalysts, primarily based on palladium and copper, is analyzed from a critical point of view. This type of coupling provides the most direct access to 1,3-diynes, which can be found in nature and are very important compounds in organic synthesis.
